What a strange question for Jesus to ask! The whole world knows that the mother of Jesus is Mary. Why would he even ask such a question? Obviously he was not asking a question of biological origin... he was asking a question of ministry connection and affinity. Who are my brethren? Who is my mother? Those who, like me, seek to follow the will of our Father.
